Biography
First Generation
Thomas Little[1] was born likely in England about 1607.[2] The first known record for him was a tax list of Plymouth Colony for 25 Mar 1633. He married in Plymouth 19 Apr 1633 to Ann Warren. Thomas was buried at Marshfield on 12 Mar 1671/2.
They had nine children (first seven born Plymouth, last two at Marshfield): Abigail (c1635), Ruth, Hannah (c1637), Patience (c1639), Mercy (c1645), Isaac (c1646), Ephraim (1650), Thomas (c1653-1676 unm.) and Samuel (c1656).
Second Generation
(Lt.) Isaac Little was born Plymouth abt 1646 and died Marshfield 24 Nov 1699, aged abt. 53 yrs. He married prob. Marshfield abt 1673 Bethiah Thomas and they had twelve children (born Marshfield): Thomas (1674), Dorothy (1676-1677), Isaac (1677/8), Bethiah (1681-b1688), infant daughter (1683-1683), Charles (1685), Abigail (c1687), Bethiah (1688-1689), Nathaniel (1690-b1715 unm.), William (1691/2), Bethiah (c1693) and Lemuel (bp 1700).
Ephraim Little was born Plymouth 17 May 1650 and died Scituate 24 Nov 1717 in 68th yr. He married prob. Marshfield or Plymouth 22 Nov 1672 Mary Sturtevant and they had nine children (born Marshfield): Anna (1673), infant (c. 1675), Ephraim (1676), Mercy (1678), David (1680/1), John (1682/3), Mary (1685-1685), Ruth (1686) and Barnabas (1691/2-1691/2).
Samuel Little was born Plymouth abt 1656 and died Bristol (now part of RI) 16 Jan 1707. He married Marshfield 18 May 1682 Sarah Gray and they had four children (born Marshfield): Thomas (1683), Sarah (1685), Samuel (1691) and Edward (bp 1698).

Previous Y-DNA testing
The Little DNA Project Subgroup: "i. MRCA unknown; Ireland; early Marshfield Massachusetts; Haplogroup R1b-L193; further testing necessary to establish lineage" lists five individuals, one whom states his Most Distant Known Paternal Ancestor (MDKPA) was Thomas Little. None of these individuals have done any Y-DNA SNP testing, but the Thomas Little descendant has done 111 Y-STR testing. Plugging in his Y-STR values into the Nevgen.org Haplogroup Predictor[1] predicts a variety of haplogroups under R1b-P312>>L21 with the top four entries (combined probability score of 95%) all falling under L21>>L513>S5668.

Further needed testing
We need additional direct male-line descendants of Thomas Little to WGS/NGS test to both confirm this prediction as well as to refine the haplogroup further and delineate descendant clades.
