First Generation
[1] Francis Sprague was born in England about 1590 and died in Duxbury in 1676. He came to New Plimoth (with his daughters Anna and Mercy) in 1623 aboard the Anne. Previous testing
The Sprague DNA Project Currently has one group stating their Most Distant Known Patrilineal Ancestor (MDKPA) was Francis Sprague. Group: "Hap'group I2a2a - Needing others to join this project or upgrade - currently unmatched with any certainty" lists one individual who claims Francis Sprague as their Y-DNA Ancestor. [1]
Additional testing needed
Additional testing is needed to determine which (if either) of these groups correctly belong to descendants of Francis Sprague. In addition NGS/WGS testing would refine the haplogroup placement.

The Sprague Project- ↑ Group Hap'group R1b - Genetic Family B also lists one individual, but Group Hap'group R1b - Needing others to join this project or upgrade - currently unmatched with any certainty lists a close STR match also claiming Francis Sprague as their MDKPA, but this line was through Lt. John Sprague (II) who was NOT the biological/genetic son of John Sprague (I).
