===Previous Y-DNA testing===
Previous Y-STR testing through the Mayflower Society<ref>https://www.familytreedna.com/public/mayflowersociety/default.aspx?section=ycolorized</ref> and the Soule surname DNA project<ref>https://www.familytreedna.com/public/GeorgeSoule/default.aspx?section=ycolorized</ref> has indicated that the ''Mayflower'' Soule lineage belongs to the Y haplogroup [https://en.wikipedia.org/wiki/Haplogroup_I-M253 I-M253], which is quite common in Northern and Northwestern Europe. More specifically, the Soule lineage can be classified within I-M253 as DF29>Z58>Z59>Z60>Z140>Z2535>S1954 (with YSC0000261 being a synonym for the "terminal" SNP, S1954). ''Y Elite'' test results may be able to place the Soule lineage more precisely and will identify a number of more recent SNP markers. Testing of multiple individuals may be able to pin individual SNP markers to specific individuals affiliated with George Soule.

Big Y testing of multiple patrilineal (all male line) descendants of George Soule has been able to refine the haplogroup/clade of the Soule famile to I-DF29>Z58>Z59>Z60>Z140>Z2535>S1954>S12289>A20032.

==Summary of findings==
+
Patrilineal descendants of George Soule have begun to identify their own subclades below A20032. All Y-DNA descendants of John (son of George) have been found to belong to a subclade of A20032>BY32573.<ref>Y-DNA descendants of three different sons of John2 all fall under the BY32573 haplogroup.</ref> All Y-DNA descendants of Nathaniel (son of George) have been found to fall under A20032>A20047/ZW20 and all descendants of Sylvanus (son of Nathaniel) have been found to fall under ZW20>A20044. All Y-DNA descendants of George (son of George) have been found to fall into A20032>A22686<ref>This clade is not revealed by Big Y-700 testing, but it is found in Whole Genome Sequence (WGS) testing from other DNA companies like YSEQ, Nebula Genomics and Dante Labs.</ref> and there is an additional sublade A22686>FGC29004.
